The Pluripotency for Organ Regeneration Research Group is looking for a Laboratory Technician to develop scientific support to the research activities of the group in the framework of the project entitled "Modelling Ataxia Telangiectasia (A-T) pathogenesis and therapeutics using human pluripotent stem cells and genetic engineering". The objective of the project is to generate ATM-defective human pluripotent stem cell lines via CRISPR/Cas9 genome editing to capture the genetic background of AT patients.
Main tasks and responsibilities:Maintenance of human pluripotent stem cells (culture, passage, frozen stocks preparation)Support on generation and characterization of ATM KO and KI cell lines from human pluripotent stem cellsGeneration and characterization of Cerebral Organoids from ATM KO and KI human pluripotent stem cells by RT-PCR, WB, immunofluorescence and confocal microscopy techniques Requirements for candidates:Essential:Degree in biology, biotechnology, or similar areas.Previous experience in cell culture and human pluripotent stem cells manipulation.Previous experience in molecular biology techniques.Competencies and skills: Excellent communication and teamworking skills, proactivity, commitment, critical and analytical thinking.High level of English.Advantageous:Previous experience in CRISPR-Cas9 genome editing and in the generation of brain organoids.Previous experience in Cloning, PCR, RT-PCR, WB, Immunofluorescence and Confocal Image Processing.Previous experience in scientific support to the research activities of international or national research labs.
